Cone and rod photoreceptor cells in the human retina have a unique sequence of differentiation. They appear to arise from the temporal and nasal retinal pigment epithelium at about 10 weeks of gestation. At the cellular level, differentiation is characterized by a series of morphological changes in the developing photoreceptors. These changes include the induction of neurofilament-68 protein and synaptic vesicle glycoprotein-2 in the outer segment, retinol and visual pigment synthesis in the outer segment, and the onset of cone cell-specific enzymes involved in the biosynthesis of the photoreceptor-specific lipids and long-chain (C30-C44) retinoid isoprenoids. These developmental changes are coordinated by specific gene expression. The homeobox transcription factor, CRX, is one of the genes that regulates these developmental processes. It is critical for development of the visual system in the mouse, and a genomic disruption in the mouse results in absence of photoreceptors and retinal degeneration. Here we report studies of the expression pattern and tissue distribution of the human CRX gene. By in situ hybridization analysis, we found CRX in cells adjacent to the retinal pigment epithelium of the human retina, corresponding to the ganglion cells and the bipolar cells in the rodent retina. In addition, we found expression of CRX in the embryonic and fetal retina. The expression of CRX was more intense in the human than in the rodent retina, suggesting possible differences in the developmental processes in the two species. These data provide the first identification and expression pattern of CRX in human retina, adding to our understanding of the molecular mechanisms controlling the differentiation of the different cell types of the human retina.Long-term monitoring of air flow directions and velocities using kinematic moiré for hot-spot detection in response to bermuda-grass proliferation in a photonic crystal.
